What is HPLC peptide testing?
HPLC stands for High-Performance Liquid Chromatography. RP-HPLC means reversed-phase HPLC, a common version used in peptide analysis.
The laboratory dissolves a sample and moves it through a column in a liquid mobile phase. Components interact differently with the stationary phase and therefore leave the column at different retention times. A detector records the response as a chromatogram with peaks.
The large main peak represents the dominant detected component under that test method. Smaller peaks can represent related substances, by-products, or other detected components.

Mass spectrometry, often shortened to MS or mass spec, measures ions by their mass-to-charge ratio. For a buyer, the important part is simpler: the test provides molecular-mass information that can be compared with the expected peptide.
If the measured result matches what is expected, the lab can report that identity is supported or conforms.
On a peptide Certificate of Analysis, the identity section may use terms such as MS, mass spectrometry, LC-MS, molecular mass, expected versus measured mass or “identity conforms.” The exact output depends on the method and instrument.
You do not need to interpret every peak in a mass spectrum. You need to see that the identity result belongs to the same named peptide and batch as the HPLC result.
Why HPLC alone is not an identity test
HPLC gives a separation picture. A dominant peak means one detected component accounts for most of the integrated area under that particular method. Retention time can support identification when compared with a qualified reference, but a main peak should not automatically be treated as uniquely identifying the labeled peptide.
That is why a buyer should not stop at “99% HPLC.” The next line to look for is the mass-spectrometry identity result.
Why mass spectrometry alone is not the whole answer

It should also contain an authorized approval rather than an unexplained screenshot of one instrument page.

Does third-party peptide testing matter?
